  • BP500-40 Bandpass Filter - Fluorescence Microscopy (Biomedical)

BP500-40 Bandpass Filter - Fluorescence Microscopy (Biomedical)

    Fluorescence Microscopy (Biomedical)

This is the most likely high-value application for an OD5 filter of this type.

    • YFP Excitation: The filter covers the 480–520 nm range, which is the perfect excitation band for Yellow Fluorescent Protein (YFP) (Excitation max ~514 nm) and related fluorophores like SYBR Green (Excitation ~497 nm).
    • CFP Emission: It can serve as an emission filter for Cyan Fluorescent Protein (CFP) (Emission max ~475–505 nm), effectively capturing the fluorescence while blocking the excitation source (if the excitation is <460 nm).
    • FITC/GFP (Widefield): While standard GFP filters are often narrower (e.g., 470/40), this filter captures the peak emission of GFP (509 nm) and FITC (519 nm).

    Recommended Light Source

    • YFP Excitation

Cyan/Green LEDs or Argon Lasers: Since the filter passes 480–520 nm, you need a source rich in this "cyan" region to efficiently excite Yellow Fluorescent Protein (YFP).

    • CFP Emission

Deep Blue or Violet LEDs (<460 nm): Here, the filter collects the signal (480–520 nm). You need a light source that is spectrally distant enough to be blocked by the filter's OD5 blocking range (specifically below 460 nm).

    • FITC/GFP (Widefield)

Standard Blue LEDs (470 nm): While this filter is wider than standard GFP emission filters, it captures the peak emission of GFP (509 nm).

    Astronomy (Visual & EAA)

    • OIII & H-Beta: The 480–520 nm range perfectly centers on the H-Beta line (486.1 nm) and the OIII lines (495.9 nm & 500.7 nm).
    • Use Case: This acts as a "Broadband Nebula Filter" (UHC type), isolating nebulae from light pollution (sodium/mercury streetlights) with high efficiency.

    Recommended Light Source

    • Emission Nebulae (H-Beta & OIII)

This filter acts as a UHC (Ultra-High Contrast) filter. It is designed to capture the specific glow of ionized gas clouds while blocking the rest of the spectrum.

    1. Doubly Ionized Oxygen (OIII)
    2. Hydrogen-Beta (H-β)

    "Broadband" Nebula Sources

Because the BP500-40 has a 40nm bandwidth (wide for a nebula filter), it captures both OIII and H-Beta simultaneously. This makes it ideal for complex nebulae that emit light in multiple chemicals.

    1. Orion Nebula (M42)
    2. Lagoon Nebula (M8)

Fequently Asked Questions

Different Types of Filter Mountings

Unmounted:

  • a bare glass optical filter

Mounted:

  • Smooth Mount for protection of the optics. No thread is there on the mount.

Metric Mount:

  • Start with M as prefix, usually seens as M25.5x0.5mm. Mostly used as camera filters

Imperial Mount:

  • Start with SM as prefix, usually seen SM1, SM05 etc... Mostly used in Optical system. (You will have a SM external thread to connecting to other objects)

C-Mount:

  • Very frequently used mounting. C-Mount is 1‘’-32TPI. And it is not the same thing as SM1, or M25.5x0.5mm.

What are the Thickness of the Optical Filters?

For Unmounted Optics:

  • 0.5mm
  • 1.1mm
  • 2.0mm

*Most of our optics are coming with 1.1mm and 2.0mm

With Standard Mounting (Mounted, No Thread):

  • 3.5mm (Default)
