  • BP532-35 Bandpass Filter - Fluorescence Microscopy

BP532-35 Bandpass Filter - Fluorescence Microscopy

    Fluorescence Microscopy

Used as an emission filter to detect GFP (Green Fluorescent Protein) or Alexa Fluor 532. The 35nm width is wide enough to capture a significant portion of the emission spectrum while the OD4 blocking keeps the background "dark."

    Recommended Light Sources

    • Blue/Cyan LED (e.g., 470nm or 488nm): High stability and longevity; provides narrow-band excitation that perfectly precedes the 532nm emission window, minimizing "bleed-through" into the detector.
    • 488nm Argon-Ion or Diode Laser: Extremely high power density and monochromaticity; allows for precise confocal imaging and ensures the OD4 blocking of the filter can effectively eliminate all excitation light for a pitch-black background.
    • Mercury or Xenon Arc Lamp: Broad spectral output; versatile for multiple fluorophores when used with a dedicated excitation filter to select the 450–490nm range.
    • White Light Laser (Supercontinuum): Tunability; allows you to fine-tune the excitation wavelength to the absolute peak of Alexa Fluor 532 to maximize the signal-to-noise ratio.
  • BP532-35 Bandpass Filter - Second Harmonic Generation (SHG)

BP532-35 Bandpass Filter - Second Harmonic Generation (SHG)

    Second Harmonic Generation (SHG)

In physics labs, it is used to isolate the 532nm signal generated when an 1064nm (Infrared) laser interacts with certain crystals.

    Recommended Light Sources

    • Pulsed Nd:YAG Laser (1064nm): High peak power; SHG is a non-linear process where efficiency is proportional to the square of the light's intensity. Pulsed lasers concentrate energy into nanosecond or picosecond bursts, drastically increasing the conversion rate from IR to green light.
    • Q-Switched Fiber Laser (1064nm): Exceptional beam quality and stability; the high spatial coherence (TEM00 mode) allows the beam to be focused to a tiny spot inside the nonlinear crystal (like KTP or LBO), which is essential for maximizing the power density required for SHG.
    • Mode-locked Ultrafast Laser (e.g., Ti:Sapphire or Yb-fiber): Extremely short pulses (femtoseconds); used for high-precision physics experiments where high temporal resolution is needed alongside the frequency-doubled signal.
    • Continuous Wave (CW) Nd:YAG Laser (with Resonant Cavity): High average power; while CW lasers have lower peak intensity than pulsed ones, placing the SHG crystal inside an external or internal resonant cavity "recycles" the light to boost the effective intensity and efficiency.

Fequently Asked Questions

What are the Thickness of the Optical Filters?

For standard mounting (mounted,non-thread):

  • 3.5mm (by default)
  • 6mm (by request)

For unmounted Optics:

  • 0.5mm
  • 1.1mm
  • 2mm

(most of the unmounted optics are having thickness of 1.1mm ~ 2mm)

What is the difference between mountings?

Unmounted:

  • a bare glass optical filter

Mounted:

  • Smooth Mount for protection of the optics. No thread is there on the mount.

Metric Mount:

  • Start with M as prefix, usually seens as M25.5x0.5mm. Mostly used as camera filters

Imperial Mount:

  • Start with SM as prefix, usually seen SM1, SM05 etc... Mostly used in Optical system. (You will have a SM external thread to connecting to other objects)

C-Mount:

  • Very frequently used mounting. C-Mount is 1‘’-32TPI. And it is not the same thing as SM1, or M25.5x0.5mm.
