BP615-8 Bandpass Filter - Time-Resolved Fluorescence (TRF) & AlphaLISA
Time-Resolved Fluorescence (TRF) & AlphaLISA
The primary application for a BP615-8 with OD6 blocking is the detection of Lanthanide luminescence, specifically Europium chelates.
- Context: Europium (Eu³⁺) is a popular fluorophore used in "Time-Resolved Fluorescence" (TRF) and assays like AlphaLISA or DELFIA. Unlike standard fluorophores, Europium has a very long fluorescence lifetime and a sharp, "atomic-like" emission peak centered at 615 nm (the ⁵D₀ → ⁷F₂ transition).
- Function: The filter isolates this narrow 615 nm peak while blocking the excitation light (which is usually in the UV or deep red) and background autofluorescence (OD6 blocking is critical here to ensure a high Signal-to-Noise ratio).
- Result: High-sensitivity detection of biological analytes (hormones, enzymes, cytokines) in drug discovery and clinical diagnostics.
Recommended Light Sources
Below are the recommended light sources depending on the specific assay type (Direct Europium excitation vs. AlphaLISA).
- Xenon Flash Lamp: High-Intensity UV Excitation (TRF): Europium has strong absorption bands in the UV range (around 320–340 nm). A Xenon flash lamp provides a high-intensity, broad-spectrum pulse that covers this UV region effectively. It is the standard standard source for TRF plate readers because the "flash" capability allows the system to wait (delay) before measuring, removing short-lived background noise while the long-lived Europium signal (615 nm) persists.
- 680nm Laser Diode: AlphaLISA Excitation: In AlphaLISA assays, the "Donor beads" are excited at 680 nm (Deep Red), causing a chemical reaction that eventually releases light at 615 nm from the "Acceptor beads." A 680 nm laser is required to penetrate the sample and excite the donor beads without directly exciting the acceptor beads or biological background. The BP615-8 filter then blocks the massive amount of 680 nm laser light (OD6 blocking is essential here) and passes only the 615 nm signal.
- UV LED (365nm or 340nm): Compact Eu³⁺ Excitation: For portable or compact fluorescence microscopes, a high-power UV LED (specifically 365 nm) can excite Europium chelates. While less intense than a Xenon lamp, LEDs offer a longer lifespan, lower heat, and instant on/off switching, which is advantageous for rapid automated inspection systems.
Fequently Asked Questions
Different Types of Filter Mountings
Unmounted:
- a bare glass optical filter
Mounted:
- Smooth Mount for protection of the optics. No thread is there on the mount.
Metric Mount:
- Start with M as prefix, usually seens as M25.5x0.5mm. Mostly used as camera filters
Imperial Mount:
- Start with SM as prefix, usually seen SM1, SM05 etc... Mostly used in Optical system. (You will have a SM external thread to connecting to other objects)
C-Mount:
- Very frequently used mounting. C-Mount is 1‘’-32TPI. And it is not the same thing as SM1, or M25.5x0.5mm.
What are the Thickness of the Optical Filters?
For Unmounted Optics:
- 0.5mm
- 1.1mm
- 2.0mm
*Most of our optics are coming with 1.1mm and 2.0mm
With Standard Mounting (Mounted, No Thread):
- 3.5mm (Default)
- 6mm (Upon Request)
