  • How it works: In multiphoton microscopy, you use a high-power infrared laser to penetrate deep into biological tissue. This laser excites fluorescent dyes in the sample, which then emit visible light.
  • The Filter's Job: You need to detect the faint visible glow (fluorescence) while blocking the blindingly bright infrared laser that acts as the excitation source. Because the excitation laser is so powerful, you need an OD6 rating (1,000,000:1 rejection ratio) to ensure none of that laser light reaches your sensitive detector (PMT or Camera).

  • For Multiphoton (Two-Photon) Microscopy, the light source is the most critical component of the entire system. Because this technique relies on "non-linear" optics (two photons hitting a molecule at the exact same femtosecond), you cannot use standard continuous lamps or diode lasers. You must use Ultrafast Pulsed Lasers.

Recommended Light Sources

The "light source" you use depends on which side of the filter you are on.

A. Sources the filter is designed to BLOCK (Excitation Sources)

These are the sources you would use to illuminate or excite your sample. The filter prevents this light from blinding the detector.

  • Ti:Sapphire Lasers (Tunable): The most common source for multiphoton microscopy, usually tuned between 760nm and 1000nm.
  • Nd:YAG Lasers: Operating at 1064nm. This filter blocks this wavelength completely.
  • NIR Diode Lasers: Common wavelengths like 785nm, 808nm, 830nm, or 980nm.

B. Sources the filter is designed to TRANSMIT (Signal Sources)

These are the sources (or signals) that will pass through the filter to be seen by your eye or camera.

  • Fluorophores (Fluorescent Dyes):DAPI / Hoechst (Blue emission ~460nm)GFP / FITC (Green emission ~510nm)RFP / mCherry / Texas Red (Red emission ~610nm)Note: It will transmit Cy5 (emission ~670nm) but will cut off dyes that emit deeper into the NIR (like Cy7).
