This is the most likely application. In fluorescence, you excite a sample with UV light, and it emits visible blue light. You need to block the dangerous/blinding UV light from reaching the detector (or your eye) while letting the faint blue fluorescence through.

  • Target Fluorophores: DAPI, Hoechst 33342, Hoechst 33258, AMCA.
  • How it works: These dyes absorb UV light (~350–360nm) and emit blue light (~450–460nm). The LP370 blocks the excitation UV (OD5 ensures dark background) but passes the blue emission signal.

Recommended Light Sources

  • UV LED 365 nm: This is the ideal partner. The LED pumps energy at 365nm (which the filter blocks), causing the sample to fluoresce at >400nm (which the filter passes).
  • UV Laser 355 nm (Nd:YAG) or 375 nm (Diode): Note: If using a 375nm laser, this filter might be too close to the laser line (cut-on is 370nm). You would need a 355nm laser for this specific filter to work effectively.
  • Mercury (Hg) Arc Lamp Broadband UV (peaks at 365nm): Traditional source for fluorescence microscopes. The filter effectively blocks the strong UV emission lines.

If you are performing Raman spectroscopy using a UV laser (e.g., 355nm), this filter serves as the Rayleigh Rejection Filter.

  • How it works: The laser hits the sample, scattering mostly laser light (Rayleigh scatter) and a tiny amount of "Raman shifted" light (longer wavelengths). The LP370 blocks the blinding 355nm laser light (OD5) but lets the Raman signal (which appears at >370nm) pass through to the spectrometer.

  • Frequency-Tripled Nd:YAG Laser: The industry-standard "workhorse" for this application is a Diode-Pumped Solid-State (DPSS) Nd:YAG Laser emitting at 355 nm.
  1. Laser Medium: Neodymium-doped Yttrium Aluminum Garnet (Nd:YAG).
  2. Mechanism: The laser generates a fundamental wavelength of 1064 nm. Through non-linear optical crystals, this frequency is tripled (Third Harmonic Generation) to produce 355 nm ultraviolet light.
