This specialized filter set is optimized for fluorophores that are excited by violet-blue light (e.g., the 445 nm laser line) and emit light in the cyan spectral region (around 480 nm). The configuration uses narrow bandpass filters for both excitation and emission, providing high spectral specificity and effectively separating the cyan signal from both violet excitation light and longer-wavelength fluorophores. This makes it an ideal choice for imaging Cyan Fluorescent Proteins and their variants, commonly used as donors in Förster Resonance Energy Transfer (FRET) experiments, as well as other spectrally similar cyan probes.
Filter Specifications:
- Excitation Filter (Ex): Bandpass 435/20 nm (transmits light from 425 nm to 445 nm)
- Emission Filter (Em): Bandpass 480/40 nm (transmits light from 460 nm to 500 nm)
- Dichroic Mirror (DM): Reflects 425-445 nm / Transmits 460-600 nm
Intended for use with common violet-excited, cyan-emitting fluorophores such as:
- Cyan Fluorescent Proteins and their enhanced variants (e.g., proteins with Ex approx 433 nm, Em approx 475 nm)
- Spectrally similar cyan dyes and probes optimized for violet excitation
