This filter set is optimized for fluorophores that are excited by blue light (e.g., the 488 nm laser line) and emit light in the yellow-green spectral region (around 540 nm). The configuration uses a moderate excitation band combined with a narrow emission band for high specificity. This filter set is particularly suitable for ratiometric measurements of intracellular pH when paired with an alternate excitation filter (such as 435 nm), as it is designed to efficiently capture the signal of pH-sensitive indicators at a physiological pH (near 7.0).

Filter Specifications:

  • Excitation Filter (Ex): Bandpass 480/40 nm (transmits light from 460 nm to 500 nm)
  • Emission Filter (Em): Bandpass 540/40 nm (transmits light from 520 nm to 560 nm)
  • Dichroic Mirror (DM): Reflects 350-500 nm / Transmits 520-650 nm

Intended for use with common blue-excited, yellow-green-emitting fluorophores such as:

  • pH-sensitive fluorescent probes (e.g., probes with Ex approx 490 nm, Em approx 535 nm)
  • Fluorescein derivatives and similar dyes when used in multi-channel applications
  • Green Fluorescent Protein (GFP) and spectrally similar variants
